Excisanin A suppresses proliferation by inhibiting hypoxiainducible factor-1α expression in human hepatocellular carcinoma cells

Purpose: To investigate the effect of excisanin A on human hepatocellular carcinoma cells as well to elucidate its mechanism action.
 Methods: Molecular docking was used determine binding characteristics HIF-1α protein. The transcriptional activation and viability were assessed using Luciferase reporter MTT assay. protein in nucleus assayed western blot immunofluorescence. VEGF mRNA levels evaluated reverse-transcription polymerase chain reaction (RT-PCR). Cell proliferation determined by flow cytometry, EdU clonogenic assays. In vivo tumor growth a murine xenograft model SKHep1 cells.
 Results: Excisanin inhibited activation, synthesis (p < 0.001). also reduced expressions addition, compound cells. model.
 Conclusion: is potent inhibitor, supporting potential development for hepatoma therapy.
